學(xué)科組介紹:魚類基因工程學(xué)科組的研究工作發(fā)端于上世紀(jì)70年代末的魚類細(xì)胞核移植研究。朱作言先生在著名實(shí)驗(yàn)胚胎學(xué)家童第周先生的領(lǐng)導(dǎo)下,通過合作研究完成鯉鯽間的細(xì)胞核移植,首次實(shí)現(xiàn)脊椎動物的種間克隆。上世紀(jì)80年代,朱作言先生在世界上首次成功進(jìn)行農(nóng)藝性狀轉(zhuǎn)基因研究,研制出世界首批轉(zhuǎn)基因魚,提出了轉(zhuǎn)基因魚形成的模型理論,開創(chuàng)魚類基因轉(zhuǎn)移研究新領(lǐng)域。以上成果作為近代中國科技領(lǐng)域的兩大重要科學(xué)技術(shù)成就被載入《世界科學(xué)年史》(A. Hellemans & B. Bunch, 美國Simon & Schuster出版社)。?
近10年來,本學(xué)科組合作培育出具有完全自主知識產(chǎn)權(quán)、生長速度快、餌料轉(zhuǎn)化率高的冠鯉和吉鯉;按照國家I類新藥的毒理學(xué)實(shí)驗(yàn)規(guī)范和實(shí)質(zhì)等同性原則,合作進(jìn)行了冠(吉)鯉的營養(yǎng)學(xué)、毒理學(xué)和致敏性研究,系統(tǒng)評價(jià)并證實(shí)冠(吉)鯉與對照鯉具有同樣的食用安全性;創(chuàng)建了“人工模擬湖泊”的生態(tài)安全實(shí)驗(yàn)體系,客觀評價(jià)了冠鯉投放到該人工湖泊后對生態(tài)環(huán)境的可能影響,發(fā)現(xiàn)在鯉魚自然分布的水域,冠鯉的生態(tài)風(fēng)險(xiǎn)不會高于對照鯉;為冠(吉)鯉的產(chǎn)業(yè)化準(zhǔn)入提供了充分的科學(xué)和材料儲備。?
此外,本學(xué)科組發(fā)現(xiàn)了魚類生長軸的信號分子在生殖發(fā)育中的作用、生殖軸/生長軸間新的調(diào)控通路及其作用機(jī)制;揭示了魚類生殖軸中重要信號分子調(diào)控性腺發(fā)育的機(jī)理;發(fā)現(xiàn)影響魚類配子減數(shù)分裂的信號分子在精子發(fā)育和成熟中的功能。首次報(bào)道了低溶氧耐受魚的成功研制。提出了魚類生殖操作育種新思路,創(chuàng)建了魚類育性可控的生殖開關(guān)新策略。?
目前,本學(xué)科組致力于經(jīng)濟(jì)魚類的分子雜交(轉(zhuǎn)基因)和分子突變(基因編輯)育種,以斑馬魚、青鳉等為模式系統(tǒng)的發(fā)育生物學(xué)研究。具體研究方向包括:魚類重要經(jīng)濟(jì)性狀相關(guān)基因或分子模塊的發(fā)掘與功能驗(yàn)證;魚類生殖發(fā)育、性別決定與分化機(jī)制解析;魚類生殖軸與生長軸之間的調(diào)控關(guān)系探索;魚類生殖開關(guān)策略的技術(shù)創(chuàng)新。?
以上研究得到了國家自然科學(xué)基金、中國科學(xué)院戰(zhàn)略性先導(dǎo)科技專項(xiàng)和湖北省自然科學(xué)基金創(chuàng)新群體基金以及科技部863計(jì)劃和973計(jì)劃等項(xiàng)目的資助。?
