魚類發(fā)育與干細胞

本學(xué)科組關(guān)注基礎(chǔ)研究和應(yīng)用基礎(chǔ)研究。一方面研究生命科學(xué)的基本問題,即受精卵如何發(fā)育成為一個完整的個體?另一方面,圍繞基礎(chǔ)科研服務(wù)于國家需求,從干細胞理論和技術(shù)出發(fā),研究重大疾病的發(fā)病機理、開展干細胞治療研究。本課題組以斑馬魚和哺乳動物多能干細胞為材料,在以下方向開展研究:脊椎動物早期胚胎發(fā)育、多能干細胞自我更新和分化機理、干細胞治療和轉(zhuǎn)化醫(yī)學(xué)等。主要研究內(nèi)容如下:?
1、以斑馬魚為模式生物,研究轉(zhuǎn)錄因子、信號通路、表觀遺傳因子以及染色質(zhì)高級結(jié)構(gòu)等調(diào)控脊椎動物早期胚胎發(fā)育、細胞命運決定以及基因表達的分子機制。為治療人類發(fā)育疾病提供機理解釋。?
2、以人和小鼠多能干細胞為材料,揭示干細胞自我更新與維持、分化的細胞及分子生物學(xué)基礎(chǔ)。鑒定調(diào)控干細胞自我更新和分化的關(guān)鍵因子;闡明其在干細胞自我更新和分化、細胞命運決定中的作用機制。?
3、干細胞治療和轉(zhuǎn)化醫(yī)學(xué)研究。研究內(nèi)容包括:干細胞理論和技術(shù)研究重大疾病發(fā)病機理;基于多能干細胞的體外類器官(類腸、類心臟等)構(gòu)建,闡明其定向分化及自組裝規(guī)律;干細胞和組織工程結(jié)合研發(fā)新型臨床替代物等等。 ?
本課題組得到國家重點研發(fā)計劃項目資助,總研究經(jīng)費約600萬元。?
